Abstract
Porous hydroxyapatite bone scaffolds containing Os-stained tissue were imaged for the first time using x-ray microcomputed tomography. Hydroxyapatite scaffolds are seeded with mouse D1ORL stem cells and cultured in vitro for 2 weeks. Samples are fixed using a multi-step OsO4 procedure in order to increase the x-ray attenuation contrast between the scaffold and the tissue. Using this fixation method, the tissue and the scaffold are distinguishable. Data are obtained in the form of an attenuation value for each volume element in the sample, and these data allow samples to be imaged in cross section in increments of 6μm without cutting the sample, or as three dimensional objects.
